农作物秸秆发酵制备丁二酸的代谢工艺优化

    该文对产丁二酸放线杆菌的发酵工艺优化,为工业规模的农作物秸秆生物转化制备丁二酸提供重要指导。采用碱解与酶解方法处理原料秸秆,并利用高效液相检测水解液,结合代谢途径的分析对发酵工艺进行了正交试验与响应曲面优化。秸秆水解液的检测表明200 g的秸秆干粉水解得约52 g的葡萄糖和约24 g的木糖。对菌株的代谢途径分析表明细胞具有五、六碳糖代谢酶系,以及在大量合成丁二酸的同时也产生少量乙酸、α-酮戊二酸等副产物。在单因素培养基础上,正交试验结果表明当主要的离子及生物素采用如下浓度(mmol/L)时:镁离子 6.0、锰离子 0.9、亚铁离子 0.6、锌离子 1.5、VB2 0.005、VB5 0.012、VB14 0.003及VB12 0.01,产丁二酸达 58 g/L。进一步的响应曲面分析表明,CO2与H2的比例、氟乙酸微量浓度、发酵pH三因素不但对产丁二酸有影响,且相互之间存在联系,当CO2与H2的比例为30.43、pH维持5.51、氟乙酸浓度为0.103 mmol/L时,丁二酸产量最高达73 g/L。代谢工艺的优化将丁二酸的产量提高了约25 g/L,使得菌株具有进一步的工业应用潜力。

    Abstract:

    It is very important to optimize the fermentation technology of Actinobacillus succinogenes for the industrial bioconversion of succinic acid from crop straw. Pentose and hexose hydrolyzed from crop straw with hydroxide and enzyme were determined by HPLC. Based on the analysis of metabolic pathway, the fermentation technology was optimized by orthogonal design and response surface methodology. Determination of the hydrolyzate showed that about 52 g glucose and 24 g xylose were hydrolyzed from 200 g crop straw. Metabolic pathway analysis for this strain showed that pentose and hexose could be transferred to succinic acid and other byproducts including acetic acid and α-ketoglutaric acid. On the basis of single factor experiment for primary culture medium ingredient, orthogonal experiment showed that 58 g/L of succinic acid was gained when the concentrations of metal ion and vitamin were as follows(mmol/L): Mg2+ 6.0, Mn2+ 0.9, Fe2+ 0.6, Zn2+ 1.5, VB2 0.005, VB5 0.012, VB14 0.003 and VB12 0.01, respectively. Further response surface analysis indicated the interrelation between those factors including the ratio of CO2 to H2, the concentration of fluoroacetate and pH value. 73 g/L of succinic acid was gained, based on the following optimum conditions: ratio of CO2 to H2 of 30.43, 0.103 mmol/L of fluoroacetate and pH 5.51. The yield of succinic acid increases by 25 g/L and the strain Actinobacillus succinogenes HF-7 has good application of industrial fermentation.
